Sometimes a tester re-raise can pick up the pot, or if you feel you have made by far the best of it, smooth call and hope they bet out again on the next card.

If a player has been supertight, you can expect to be up against a stronger hand. This and betting patterns should be more than enough to form an opinion of whether an opponent is bluffing or not.
